Forskolin Extract

Forskolin Extract is produced from Coleus forskohlii, a botanical that contains a unique substance known as forskolin. Forskolin, a diterpene, activates the enzyme adenylate cyclase, which results in increased cyclic adenosine monophosphate (cAMP) in cells. Forskolin's potentiation of cAMP enhances cardiovascular health by supporting healthy blood pressure levels already within the normal range, promoting a healthy coagulation response, and enhancing heart function.

INTRODUCTION
The cardiovascular system consists of three interrelated components: blood, the heart, and blood vessels. Blood transports various substances and helps regulate several life processes. The function of the heart is to continuously pump blood throughout the body. To accomplish this, the heart beats approximately 100,000 times every day. Blood vessels provide structures for the flow of blood to and from the heart and the exchange of nutrients and wastes in tissues. The also play an important role in adjusting the velocity and volume of blood flow.1
The wall of a blood vessel generally consists of three layers, or tunics, of different tissues. These layers include an epithelial inner lining, a middle layer consisting of smooth muscle and elastic connective tissue, and a connective tissue outer covering. The middle layer, also referred to as the tunica media, is the most variable of the tunics. The primary role of the smooth muscles located in the tunica media, which extend circularly around the lumen like a ring encircles a finger, is to regulate the diameter of the lumen. The rate of blood flow through different parts of the body is regulated by the extent of smooth muscle contraction in the walls of particular vessels. The extent of smooth muscle contraction in blood vessels is crucial in the regulation of blood pressure.1

HOW DOES IT WORK?
Coleus forskohlii is a botanical that has been used in Hindu and Ayurvedic traditional medicine since ancient times. Forskolin, the active constituent derived from the root portion of the Coleus Forskohlii plant has been extensively studied in the laboratory setting. Its mechanism of action has been found to be the activation of the enzyme adenylate cyclase, which results in increased cyclic adenosine monophosphate (cAMP) in cells.
cAMP belongs to a class of substances known as “second messengers,” and is one of the most important cell-regulating compounds. Hormones and neurotransmitters also activate adenylate cyclase—but forskolin appears to be able to activate adenylate cyclase by itself. Thus, forskolin can increase cAMP without the assistance of hormones or neurotransmitters.2,3,4
As mentioned before, cAMP is called a “second messenger” because it can receive messages from outside the cell and then transmit them within the cell, where it activates numerous other enzymes involved in diverse cellular functions. The increase in cAMP levels caused by forskolin provides support of cardiovascular health through multiple mechanisms.
Forskolin has been shown to support healthy circulation and coagulation factors in both animal and clinical studies.5,6,7,8 One study investigated the effect forskolin had on hemodynamic (blood flow) functions. Results were measured by invasive techniques using the thermodilution catheter method. Forskolin was found to dose-dependently enhance hemodynamic functions via healthy vasodilation.9 Forskolin's ability to enhance healthy blood coagulation factors also supports cardiovascular health. One animal model study investigated overall platelet metabolism, measured by a microcalorimetric technique after treatment with forskolin; it was found to be significantly enhanced.8
Studies have also demonstrated that forskolin, via cAMP potentiation, also enhances healthy heart muscle function.10

Other Applications
In addition to the promotion of relaxation of smooth muscle tissue and healthy blood circulation, forskolin has been shown to support healthy eye health, most likely through cAMP activation. Both animal and human studies have demonstrated forskolin's ability to support healthy intraocular pressure.11,12 Forskolin, via activation of cAMP and subsequent vasodilation, has also been shown to support respiratory health.13,14
